It had something to do with a setting in vista - it is mentioned in the link above. Maybe the vista update changed one of this settings.

I have a problem with a brand new Sony Laptop. Itunes doesn't save the library after adding folders and restarting the program.
I was searching for a solution and found this: http://www.ipod-forum.de/i
It worked to fix it with this sony hint - but after a windows vista update (some days ago) the problem is back.
That's what i did. Start itunes - add folders to the library - the music is there - close itunes - start itunes - the library is empty.

Some ideas
Vista is known for it's security implementation, it su..., the user that you used to login most likely has no rigths on "My Music" or "iTunes Music" folder so if you let iTunes manage your library it can't move the added music to the "iTunes Music" folder.
A work around could be manually copying your music to the "iTunes Music" folder
Another popsibilty could be that the folder coantaining your music is read only.
At the end I think your problem is related to permissions on the originating folders or the destination folders.
Check them to see their properties
