Yes, but you should do so cautiously. Depending on the reliability of the connection between the client and the host you do put the stuff you're editing at risk, e.g. we have seen scripts, tables and layouts disappear if a connection is interrupted while that item is being edited over an intermittent IP connection (e.g., over the WAN).
skeleton key is correct, but more than that you can corrupt the entire database file on the server if the connection is slow or intermittent, especially if the FileMaker file is large and you are making changes to the structure. Making changes to layouts and scripts appears to be much safer than making changes to the database structure such as tables, fields and relationships.
A safer way to work remotedly, especially if the network is in question or the file size is large, is to use Terminal Services, LogMeIn or some similar service to access the file within the network. This has worked for us in the situation above because it means we're operating and saving locally.
