My experience has been that one tunes the O/S, then the RDBMS / database, then the SQL statement. At the statement level, it's a popular idea to correct those events that contribute the longest wait time.
That said, there are far better sources than our volunteer comments, given so broad a question. My High-Performance Tuning with STATSPACK book runs about 700 pages, for example.
