Which version if exchange are you using?
Is the java programme running on a different server?
Is the programme authenticating with your SMTP service? I.e is it supplying a valid domain username and password?
In exchange 2003 you may need to add the IP address to the allowed relay section of the default virtual smtp server located in Exchange System Manager > Administrative Groups > First Administrative Group > Servers > Servernsme > Protocols > SMTP
in Exchange 2007 it's in Exchange Management Console > Server Configuration > Client Access > Send Connector tab you need to add the IP to the client connector.
