are you using SAN or standard IDE/SATA drive. If its SAN then there are different ways to improve performance, If its standard HDD then there are different.
In my opinion you probaly need, defragmentation, check IO bottlenecks, HBA cards/ RAID config

more memory?
Do you run advanced server or just standard server?
Advanced server will allow more memory, and so reduce disk thrashing.
Also, do you have raid? Can you add more drives, that will make things quicker..
Optimization of the server can also help. like page file, etc.
database may need optimizing and defragmentation, as does the filesystem..
running out of diskspace?
An increase in clients? Addition loads like new software or backups, virus protection etc...
