Here are some good sites that I've found for audio. I know next to nothing about PC video, so I won't even try to point you in the right direction.
I tried, without success, to turn my Athlon 700 box into a PC-DAW (digital audio workstation), but eventually, I built a system from scratch using an ASUS P4B533-E motherboard, Maxtor drives (one for OS and apps and the other just for Audio), and Echo Audio's Layla24 audio interface. Building a PC from scratch is a great exercise. The hardware is the easy part... getting the BIOS and drivers to do what you want is the hard part, but with a little effort, you can build a system for $2000 that would cost you $4000 if it was built for you.
At first, I thought that I only had to find the right sound card, but learned that the "chipset" (in the case of the P4B533-E mobo, it's the Intel 845E chipset, which you'll see, as you read these websites, is one of the two or three that are the most compatible when it comes to audio) is the most important thing when it comes to audio on a PC. I'm talking professional-level multitrack audio recording, here, not game or DVD audio, mind you.
Memory speed is an important factor and I have only found one source for CAS 2-2-2 memory for the P4B533 mobo: www.mushkin.com. Expensive, but worth it. I've read not to get ECC memory because, although it is more stable than regular memory, it is slower, and you can't get CAS 2-2-2 memory with ECC (at least you couldn't three months ago when I built my machine).
