You just plug in your fluke toner to a network jack in the office and switch it on so it is generating tone.
Then press and hold the button on your probe and touch it inside the ports in your patch panel one at a time until you can hear the tone.
You can test it first by switching on the toner, pressing the button on the probe and placing it on the toner cable, you should hear the tone.

You'd be better served by buying a tone trace kit, putting the tone generator into one wall jack and testing at the patch panel end. Progressive Electronics makes a model I've been very happy with.
