For some reason users here can select "Open Other Users Folder" in the File menu of Outlook and type in anyones name and open their Inbox - not very secure!
I can't find a way to disable it.

Read permissions means that the users can read the permissions of the object - that is not the mailbox itself, but the object - the account.
Undo that deny, as it can cause you a problem.
Server side, there are only two sets of permissions that can grant access.
The combination of Send As / Receive As and Full Mailbox Access.
Rule number one though - do NOT change any permissions unless you 100% sure what they do. The Exchange permissions are complex and it is easy to lock yourself out of the mailboxes.
Most likely permissions has been granted to a group that everyone is a member of. You need to find it then start to look where it has been granted.
Be careful.
Finally, Exchange caches permissions. Therefore if you make a change, do not expect it to block access immediately. It can take two hours before a change is effect.

You need to look at the permissions and make sure that only the user has permission to the mailbox. If there are exceptions, where assistants should have access to certain areas (calendar, contacts) of their bosses mailbox then they should be delegated accordingly.
