SQL Server problem with Practive management software

We have a server runnimg microsoft sql server which is are practice management server. Over the last feww weeks for no apparent reason that I can see a uers will try to log on to the practice management software  and it will say"connection open or databse unavabable access is denied" And then maybe an hour later it will work this is happending with random users and reandom computers any thoughts to what is happening/
