Query Performance

we are about to start a new project Using SQLServer 2005, VB.Net 2005 windows application.

One particular User requirement is as follows.
within the system there is one screen which the user wants to refresh every 5 seconds. the screen is simple windows with a gridview displaying data from a view. the view will have a maximum of 6000 records. at the startup they can choose to display all or add few conditions, so the number of records displayed on the grid varies.

now this application runs on about 80-100 terminals.

so from 80-100 terminals every 5 seconds the query fetching the data is ececuted.
to check the performance we made a sample application and we started testing it on 80 terminals.
upto 10 terminals the behaviour seems to be OK. above 10 terminals the query response times are increased and sometimes its taking about 1 minute to refresh the screen.

when we execute the query from SSMS it returns the data within 1second.

now i need your advice on what counters to check and how to make it fast.
