Microsoft, Access, 2003, some users get "don't have exclusive access" message

I've got two separate groups of users with Access 2003, using two separate databases (mde files, run from a network drive). Both have been experiencing an odd problem. One of the users must be the first to open the mde or they get messages about not having exclusive access to the database. Sometimes they are able to get in after clicking the OK button a lot; sometimes they can't. If others get out and allow them to get in, they are able to get in and the others can get back in. One of the database programs this occurs with has been in use for years, with no hardware changes or user changes, but this problem just started occurring recently. One program uses Access as the back end; the other uses SQL.

I suspected Windows updates, but if that were true, I should be seeing this happen all over the building. (We have about 50 active Access databases that run as mde's from a network drive and are shared in this way.) I read a similar problem answered on this site that claimed mde's are just like that, but my 10 years experience with Access says this has worked in the past for us.

We also attempted to swap out the pc for one of the users with this problem. It seemed to fix things, then she left and the user who took her place now gets the same error with the new pc! The users all have read/write permission to the database program's folder and the data area. We're at the hair-tearing stage, so any help will be appreciated.
