can partitians help speed up startup and programs

can partitians help speed up startup and programs?

this is what i have
C: Main Drive 500GB
installed is Operating System, Main Custom Database Program, Documents & Storage 10 users

my thought is partitian single C: Main drive as follows:
C: Operating System 25GB
D: Program 100GB only requires 25GB
E: Documents User 1 25GB
F: Documents User 2 25GB [ continue this for each user totaling 250GB ]
Z: UnAllocated 125GB for future use

this is on a Windows Server 2003 system.
my thought is that each partitian will make the server response quicker to each user.
everyone uses the same D: Program but will have their own map to the network for a roaming profile which i thought i could do as a partitian for each to reduce queries to all the same drives?
