if you're using routing protocols, then you'd just add the network statements on each end. The ASA and the routers just need to advertise what's behind them. Since you're using all cisco equipment EIGRP would be an obvious and easy setup. Only concern yourself with the private networks.
If you're using static, you have to add static routes at each end so the router knows about network that it's not attached to. Again, only concern yourself with the private addresses.
You still need a default route to the internet, but aside from that, it's routing as usual.

Are you trying to implement something like DMVPN or do you just want your two spokes to go through the hub like a point to point setup?
