Actually, if this is a Linux or Unix system, you can run the VM as root. Then simply revoke access to the device node in /dev from everyone. Of course root will still be able to access it, but other users will not be able to mount it at all. The only drawback to this approach is that you would have to run the VM using the sudo command with the root password.

There is no way to do this. If the drive is plugged, the OS can see it (otherwise, the VM wouldn;t be able to use it).
