Writing on ISO image
I mounted an ISO image using "-o loop,rw" option.
Then I went into the mounted folder and tried to make a slight change into one of the files. It appears to be still readonly.
What is the simplest way to modify a inside an ISO. Is there some option like tar does, where I remove that particular file from the ISO and then make changes and add it back to ISO or do I need to extract ISO and make changes and remake it? Please give me step by step process.

AFAIK you should mount -o loop then copy the intended dirs/files you want to change to another location, change them and then recrate the ISO image.
