Users unable to acces server

I'm getting an Event ID 1000 error in the app log stating unable to access registry key in sysvol.
The users are unable to access any resources on the server. It is a single server environment. When I expand domain controllers in ADUC - properties - group policy it states that a domain controller was not found.

These types of errors are usually due to DNS problems.

Are the client machines pointing to the domain controller (and ONLY the domain controller) for DNS?
