go to computer management->shared folders->open files and make sure that no one is accessing the file.Its always recommended to copy the profiles after hours.
You can use windows NT backup to copy and restore the profiles ( can copy the open files)
or use
robocopy source destination /Copyall

This is very simple issue. OS has generated a simple and straight message which indicates that Media Player.lnk is being used by some process or user. Make sure:
1. This profile is not being used by any user while copying. DIsconnect all the sessions and then try again.
2. Media Player application must be closed.
3. Try to find out the handle/dll which is locking Media Player.lnk. use Process Explorer. Download it from net. Run Process Explorer on both: local and remote computer - just to make sure it is not being used while copying.
