Printer definitions are all gone from Windows 2003 SP2 print server. DEP throws Spooler SubSystem App has been closed a great number of times.
This morning my Windows 2003 SP2 Printer server was unresponsive to the keyboard and mouse.
When I rebooted it I get a long series of 9 DEP SPooler Subsystem App errors and none of the 37 printers are listed.
If I try to add a printer I get 'Operation could not be completed. Printer spooler not running.', even though if I look in Services.msc I can see the spooler is running. If I try to restart it .
What could effect ALL defined printers at once?

Thank you for this! Sadly, not knowing what else to do and in dire straits indeed, I rebuilt the server last night and upgraded it to 2008.

I solved the issue myself.
you will need to clean the spooler, you can find the required files at the location below.
http://honestpchelp.com/2011/cleanspl-exe-print-spooler-cleaner/
Extract the zip file, you will see Cleanspl.exe and PRINT_DEFAULT.reg
Run the Cleanspl.exe and click "Clean Spooler" and click Yes to Start.
Click Yes on each notification and restart your computer.
When computer boots up, the print sub-system will be cleaned-up.
Double click PRINT_DEFAULT.reg to restore default Printer Monitor.
Restart the server
