As you 100 % satisfaction is impossible to any experts. We will try our level best to full fill your question. In my first commet I was already mentioned the infopath applications are also useful for e leaves. Why can't give points to first comment, and coming to other comments, I tried to go in a manner, if you are not capable of holding licenses of Infopath, then choose for free template.

If you are familiar with Infopath then you can build the application in the Infopath and use Infopath form services in MOSS 2007 and do some customizations / Create some workflow using SharePoint designer 2007.
Or
You can use AbsenceVacationSchedule server template. You can customizations. I was not done practically. I'm giving some outline guidelines for starting your applications.
We created our own structure in Dot net, So we not going for SharePoint for this type of requirement.
