i have just started to use Designer, and have been making use of the Data View and Data Form web parts extensively. i've been able to create customizations that were not possible outside of Designer.
i am the only one in my company who uses Designer. the rest of my users can't even manage to use the GUI interface of sharepoint for adding web parts. there's no way i'd let them use Designer.

I use sharepoint designer quite often, making workflows, changing template designs, changing CSS, backing up sites, etc... all wrapped around microsoft office sharepoint server.
I have not yet implemented sharepoint 2010,l but I'm assuming all custom webparts and workflows should work fine and connect right to sharepoint 2010
I do not let my end users use sharepoint designer, as there is a high risk of error, I would say this is more of a manger / admin tool for making workflows and custom parts. My end users use sharepoint server as a daily intranet, they collaborate and organize information daily.
