OK, stopping the Microsoft ISA service within services on the server, just changes the Internet Explorer error message to 'Internet Explorer Cannot Display the Website' on the client PC's.
For reference, if a client PC is logged on as administrator ( with ISA active ) than the Internet works fine.
With 2 network cards presumably ISA is also providing the routing to the Internet, therefore needs to be active!?
Is there another option that can be tried to allow Internet Access for all users, or to block no port 80 outbound traffic.

For whatever reason, your Internet Security (and Acceleration) server is blocking the access. It's like Websense or Surfguard. A request goes out (through the domain->isa server) and is denied based on a ruleset (which may be blank or deny all).
It seems like you aren't using ISA server, so try disabling it, or removing the role from the domain controller. otherwise, you can go to the ISA Settings and try and fix the configuration, but it can get FUBAR'd pretty easily.
If you don't need it, best option is to disable/remove it. Microsoft ISA Server
