You can try playing around with Microsoft Management Console.
Control Panel | Admin Tools | Local Security Settings | Software Restriction Policies | Additional Rules. Then right click and add a new path rule.
But remember, clever users will just copy the program to the hard drive.

You could could disable the floppy, CD rom and usb port from the motherboard bios.
Hit the 'Delete' key when prompted during the boot to access the bios.
