I suppose you're trying to run the defrag by clicking on the shortcut in the Start Menu? Have you tried this: Start -> Run -> type defrg.msc and hit the Enter key.

Also, you can try reinstalling it. Download and open Dial-a-fix (http://wiki.djlizard.net/
Try going to Start, Run, and type chkdsk /r, hit enter. You should get a message saying that it can not run because the volume is in use, would you like to schedule a chkdsk at the next system startup?
Say yes. Reboot and it will run a chkdsk before goign into windows.
After chkdsk finishes, try defrag again
start first by checking the free space on the disk -15% minimum - and free up as many space as you can.
then boot into safe mode (hit F8) and run defrag fram there
if that does not work, run sfc /Scannow from the run box, or even a repair install :

try from command prompt...
defrag c: -f
the -f makes the drive defrag even if its below the diskspace threshold.
Otherwise, try running a windows repair if you have the CD?
