If the file is residing on a windows server, sometimes the server thinks the file is still be accessed, but it can literally have days go by with no activity. When a user reports something like this happening, I will log onto the Windows Server the drive is phycially at, right click on "My Computer" and go to "Manage" This alows me to click on "Network Shares" and see what files are currently being accessed. Under "Open Files" I find the file the user states they are having problems with and right click on the file and choose "close file" Theis allows the file to be opened up for use again.

As an alternative, you may reboot the drive.
