Buliding a web site ready for "Load-Balancing / Web Cluster" environnement?

Hello, i'm currently starting a new web base project for a medium business. The web site will be in ASP.NET 3.5 and SQL 2005. They need to host the web application on a "Load-Balancing / Web Cluster" environnement. (2 x Load Balancing server, 2 x Front-End server, 2 x SQL/Miroiring Server).

Should I write my code and organize ASP.NET application differently then i does normaly for a normal web site with only 1 server.

I heard about spliting my code with business logic/layers....but can't find  what does it really mean when developping.

What does it' mean exactly and what i should care about when developping my application.

If some one can provide a "really basic" exemple of what does it mean.. i will appreciate.
