With so many solutions, how can you tell which solutions are most likely to help you and which ones are not? Need info on Application Server-Based Development VS Traditional Locally Deployed Apps

Can someone please provide me with some info (or some links) on issues involved with Developing Application Server-Based Applications VS Traditional Locally Installed/Deployed Apps?  I know this can get very complicated, but for now I am just looking for some bullet-point type facts...primarily from the application development perspective.  

What does it mean to me as a developer if I am writing Java or .Net, for example, to be deployed each of the 2 ways mentioned above?

I know this is a "loaded" question, but any insight for this beginner will be much appreciated!
