Windows 2003 Password policy implementation

I am about to implement a password policy on a domain, where there has been none in the past.

I am going to implement a minimum of 6 characters, 8 password history, complex passwords enabled, 90 day rotation.

My question is, when I first enable this will I have to wait 90 days until the users are asked to change their passwords?

Is it best to set the rotation to 1 day initially and then change it to 90 days after that? this assumes that all users will log in that one day, which might not necessarily happen.

What would be best practice in how to go about this implementation?

It seems that i must implement this at the domain level and cannot test the policy on an organisation unit containing test machines.. is this correct?

Many questions, deadline approaching.. any help would be appreciated
