All the bulk delete tools will remove legitimate email. There is nothing that can remove just the spam.

You would have to stop SMTP on both servers, so no legitimate email comes in, shut the frontend server off the from the internet and then clean it up.

There is no way of preventing spam. Your server will get spam. You haven't said what kind of spam you have. If it was NDR spam (postmaster) then recipient filtering stops that, if it was open relay, then you should close that off. My spam cleanup article goes through both of those.
The best antispam software is the one that works for you. There is no one size fits all. Everyone will have their favourite, but that will apply to what they have seen on maybe a few sites. As someone who has worked on 100s of sites, I have seen all of the major products work successfully and seen them work so poorly they had to be removed. You have to evaluate the different products to see what works for your individual email profile.
