How can one stop spams coming in

Like alot of internet users I suffer from a high number of spams entering my email box. I have Windows Vista which is effective in filering the spams into the junk mail but I still have to go through them to pick out the odd email which is not spam.

Alot of the spams are promoting the same websites but do come from the website and cannot be replied to. Is there any way I can stop spams coming in at all?
