I have a dropdown, referenced by cell M20 (not object), with data validatation source of =$K$52:$K$59
Now, I want to change the source data validation in code if a checkbox value is checked:
Private Sub chkTarget_Click()
Call setTargetValue 'set blnTarget based on value of checkbox
If blnTarget = True Then
'WHAT GOES HERE TO CHANGE SOURCE TO CELLS K56:K59
end if
End Sub

That's very strange. I actually recorded that from the macro recorder, cleaned it up and tested it. (Using Excel 2003)
Are you using a lower version of Excel? I didn't check to see if all the objects exist in prior versions.
Other thing you could try is putting the above all on one line, and avoiding the continuation character:
.Add Type:=xlValidateList, AlertStyle:=xlValidAlertSt
