Using a user form in Excel, i have several fields. To simplify my problem i will assume just 1 field. 'Location' field. This is a combo box (or listbox) thats populated when form is opened from stored values in Access db (or can be sheet 5). When user starts to type in combo the field is auto completed. if the value doesn't exist its automatically updated to database.

Example data in database. Alaska
                                       Alabamha
                                       Athens
Here when the user types At, the field is populated with athens, or when alab is typed its populated with Alabamha.
Now if the user types Aus for Austrailia, which is not in list, the user can be propmted to allow form to update db or automatically do it.
Whats required !
a user enters 'Austrailia' - the form prompts to add value. in backgroung the database is opened, the table is opened and a query or some check is done to ensure the value doesn't exist. If not it adds the value , closes the table, closes the database. And this happens without the user seeing access opening.
Although if the value isn't populated it would indicate that the value is not there, The checking to compare and see if a particular value exists is IMPORTANT.
