Not sure about Premier specifically, but usually when looking for lock files I'd sort the folder's contents by date (Detail view, then click the Date column header) and look for files modified most recently. With MYOB that usually shows loads of files modified a few years ago and then 2-3 files modified recently. Your data file is obviously one of them, and then anything else just a few bytes long will just be the lock file.
If it's a permission problem - which version of WIndows are you running and where is the data file? MYOB has a habit of putting your data file in the program directory. Try moving your data file into My Documents, then open MYOB and click Open Company and browse to My Documents.
If that works, you can later create a shortcut to myob.exe followed by a space followed by your data file (eg C:\MYOB\myob.exe "C:\Documents and Settings\GCTIT\My Documents\myob.dat") - using "quotes" around either file path if it's a long file name and that'll cut out the need to always Browse.
