If you are using Photoshop 7.0 or CS, got to Window in the menu bar and select info. This should bring up a palette. At the bottom of the palette, the X and Y coordinates of the cursor are show. The Width and height of any selections are also shown.
You can also user the Rulers CTRL + R to help out.
Click in the top left of the rulers, and drag to set the 0 point. If you click in either the h or v ruler, you can drag out guidelines. You can snap to these guidelines.
Finally, if you need to change the units of the coordinates from measuring units to pixels, you can do that in the preferences:
Edit > Preferences > Grid & Rulers
