I need to redue a 36MB TIF to something useful for my main webpage.
1) how do I reduce the file size without shrinking the image area?
2) what's the best screen size for an image which will take up my whole screen?
3) what's the best file format?

1. To reduce the file size without reducing the image resolution (area) you need to use a file format that compresses the image.
2. The best size for an image to take up your whole screen depends on you monitor resolution.
3. The best file format depends on what it will be used for. JPG is very common and works for websites and compresses for smaller size. PNG appears to be popular for websites as well. GIF works for websites also.

rlandquist is right, but may I add that you are going to want to reduce the size of that image. Right now it is about 3300x3800, that's (way) too big for web use. Most (read most modern) browsers will scale the image down to fit in the browser window, but even if you compress a 36mb tif to jpg, you're still looking at something a bit large for web. A size recommendation would be arbitrary on my end, but unless it's necessary for the user to be able to click on the image to enlarge the image (a lot) to look at close detail, I wouldn't recommend anything larger than 1920x1080. That's the top end of displays. With a jpeg setting of 90 that will yield a file that is about 1.5mb in size.
If you mean enlarge the image when it is clicked...
Just leave it at the size it is, and compress it (save as jpg). It will take longer to load and use more bandwidth though. Make sure when you put it in your webpage, you specify the width and height that you want it to initially be displayed as though. Like this:
