C:\Windows\System32\spool\
And then reboot.

To delete a printer from your system:
1 - Open 'Control Panel'
2 - Open 'Printers'
3 - Right-click on the icon for the printer to be deleted
4 - Click 'Delete' on the menu that appears
You may need to disconnect the printer from the machine for this to be successful.
