This is a hefty server I'm talking about here, the load is fine and the network card is under utilized. The network connection on the server is fine and i can access all resources on the network from that side. Pings are successful to the server from all areas of the network that I tried. I was only trying to access the C$ share, however remote desktop works fine to the server in question. There has been no hardware changes on it at all.
It was the end of the day when this come about, so I aint done much in the way of troubleshooting as of yet, but I did enable NETBIOS over TCP/IP on my network card on my PC and that fixed it on my machine. I know that that is the protocol used when accessing shares in this way, the funny thing is we dont disable netbios via DHCP anyway and that is the only machine it is happening with.
There are no firewalls on the machine and I disabled the anti virus.

Were gonna need more information.
Whats the load on the server? Is the CPU spiked?
How about the network connection on both the server and client?
Do pings to the server fail?
Can you access other resources on the same server?
Have you replaced network cables or network equipment? Possibly faulty hardware?
This could be a lot of different things please tell us what troubleshooting steps you have tried so far
