1) 100 mbps A-B | 20 mbps HUB to B
2) 1/2 mile A-B | Few miles from HUB to B
3) 2000 users in every spoke | 5000 in HUB
4) Replication is open 24/7 at 30-60 minute intervals (Currently)
5) Lots of changes happen at spokes as well as HUB.
I'm not looking for formulas as much as "best practice" Do we keep our HUB to Spoke layout over a 20mbps llink from HUB or start connecting spokes together due to 100mbps links between those? Does the single physical 20mbps connection to these spokes make a difference in the decision? 20mbps is still very fast for AD replication correct. Currently no queues are building up that I know of.

The replication architect depends on no of factors.
Link speed
Distance between the sites.
No of user in a sites.
Mostly in enterprise replication is scheduled in right time to preserve the bandwidth.
If you make lot of changes in AD or users travel frequently you can have replication configure on 15min basis.
The replication you can configure to have from nearest site to get update from nearest dc.
