IIS 5 Using URL to Choose Server

Hello Experts,

I currently have 2 IIS 2000 web servers and would like to set it up so when a user types in a specific URL or link it directs traffic to an application run on the secondary server.
DNS is handled in house.

I have one IP address and do not want to use a port number in the URL.

I know you can use HTTP Headers to direct traffic to multiple sites on the same server.
Can you jump servers or do i have to make a changes to DNS maybe even my router?

Example

http://www.mysite.com/application1... = IIS Server 1
http://www.mysite.com/application2... = IIS Server 2

I want to make sure the link includes my domain name.
