I changed the physical files location, and was able to still access the page and view the IIS7 logo. I added another directory (phpmyadmin) which uses PHP files. When I attempt to access that directory I am getting 403 Access Denied. This is the same case for the second website which I am attempted to create from scratch.

It seems like you dont have default page ( normally index.php for phpmyadmin files) setup for phpmyadmin site/virtual directory. Setting up same in IIS would fix this for you.
