I do it all of the time with no domain assigned. In fact, not only have I used my Vista Ultimate PC and Vista Basic laptop successfully, I stuck PCA 12.1 on Windows Server 2008 RC0 and it connects up with no problem to a WinXP PC with PCA 11.5 on it.
I suppose it's not a case sensitivity issue with the login and password? Have you checked that?

Your tag mentions PCA v12.x so that's good. 11.x does not work on Vista.
How are your firewalls configured (particularly on host)? I've had that block PCA connection requests.
While you're at it, make sure callers name and password are configured correctly on host computer.
