So as I understand it you already have the VPN configured between the 2 sites. to test connectivity you should be able to ping the remote gateways internal IP address from teh command line on any of the PCs in both locations. If you can't then we need to troubleshoot the configuration. If it's setup correctly you should be able to rdp to a remote IP address or connect to a windows share by going to the command line and typing \\10.10.10.10 for example.
Which applications are you trying to make work? Usually you need to use IP addresses instead of computer name unless you have all of the PCs on both networks pointed to the same or associated DNS servers.

If you have a router to router based VPN connection, you should be able to connect to the other side straight through without the need to do anything on your side. Have you tried accessing the other network resources already? Did you receive any error messages?
