We have 2 global sites in the US and UK, with a MPLS link between the two. It seems that when the people at the houston site are logging on to their computers (~7:30-8:30am) the users at the UK site when trying to access data on the US servers experience major delays. We saw this in action by observing ping times jump from 150ms to close to 1000ms from the UK to US. However, ping times from US to the UK site over the same VPN look good.
I am not sure the users logging on has anything to do with the issue but it might be relevant. We have AD, DNS, DFS and WINS replication between both sites but not at a specific window. There are static "helper addresses" specified (in the cisco routers) between sites to aid with NETBIOS broadcasts.

It can be very helpful to compare the ping times for both the wan and lan interfaces as suggested above. You should probably manually set the packet size to simulate real traffic. If you're seeing substantially different ping times on the wan and lan side then you can try traceroutes using d3tr to get an idea what you're looking at. Have you confirmed to make sure that your connection isn't saturated at these times?
The WAN and LAN ping times were very different. We determined it was a massive amount of intermittent traffic that was causing the problem from the Houston side. We checked top talkers and turned off some DFS replication we did not need. Waiting to see if it made a difference.

It's a 3d traceroute program. It can be very helpful when it comes to finding out where a speed/connectivity problem is. www.d3tr.de
Hopefully adjusting your DFS replication helps.

Which devices are you pinging.
Do you get a different response when you ping the Houston router WAN, LAN and server interface from the UK?
