You cannot use serial port through ESXi. If you want to use serial port then you have to install ESX4

Yes it is possible, our office use VM for door access system controlled via the serial port
Just add the serial port to your VM
Within the guest OS, you can communicate to the serial port directly
I've given similar answer to similar Q before, i'll try to find it if can help you further
