The short answer is No, you can't prevent a user from modifying the file. Marking it as read-only simply generates a message when you attempt to change it -- a simple click on the message will allow changes with no problem.
Doesn't help with Virtual PC, but one alternative that would do what you're asking is to use VMWare instead to create the virtual machine ... and to distribute it to the users with the free VMWare Player application ==> this only runs the virtual machines, and does not allow any configuration changes. Works very nicely -- and although VMWare Workstation isn't free, VMWare Server is, and it can be used to create the virtual machines with no problem.

you could mark the .vmc file as read only in the properties of the file
this way it can't be edited.
