If link-building is not measurable, then why bother putting ALL this time in the hope that SOME DAY I MIGHT (or might not) get listed in top 10-20 search results??? I am confused I guess.

Your question was about "a backlink" not about "link-building" and as I said you can't measure the effect of one backlink.
You can however measure the effect of link-building or to be more precise 'observe' the effect of link-building. The number of backlinks, their quality and the anchor text will all have an effect. In general an increase in the number of on-topic quality backlinks with keywords in the anchor text will improve a site's position in the SERPs for those keywords.
Although backlnks (and title) are probably the most important elements there are over a hundred others in Google's algorithm which is why it is not possible to measure the effect of adding one backlink but only to observe the effect of adding many.

You can't.

It does not have an effect because Alexa rankings are based on the usage patterns of Alexa Toolbar users only.
