I'm using the OLEauto in one of my Delphi applications and have no problems in CreateOLEObject('Excel.Application'), but I want to open an Excel XLS file and place the content of Edit1.Text in cell B4. How do I do?

Try this. I didn't check it:
with CreateOLEObject('Excel.Application') do
begin
  Workbooks.Open('C:\somepath\something.xls');
  Range('B4').FormulaR1C1:=Edit1.Text;
end;
