What, specifically is the file you are trying to delete?  Depending on what the file is, it may very well be in use by the system, or a service running on the system.

Cybertoddicus is right it probably is in use by something.  A couple of things you might try all of these have worked for me when I've had the same prob.
1)Boot into safe mode and try.
2)Kill all services that you don't need and try.
3)Logon as seperate user and try

Could you provide more info on the file you are trying to delete (i.e. the name of the file, what software it belongs to)?
