1. How can I set "vacation message" in case user is not in the company ?

2. What is the MAIL or RCPT format when I telnet to X-Change on port 25  and use this command for test sending mail.?

3. Once telnet to X-Change port 25 , and type "help" , I see NOOP, AUTH, HELO. What do these mean ?

1.  So you want an autoreply if the user has left the company?  Probably the best way at least initially is to set up a rule on the person's mailbox to autoreply with a specific template.  To create the template, open an email message, type the text you want, and save it somewhere on the file system.  Then set up your rule.

You can also use an Out of Office.  

If you want to do it on the server side, you could use na Exchange event script.
