I am writing a POS program in vb6. My program needs to response when the cash draw has been closed. I want to know how to capture the "draw-close" event by VB6.

you need some kind of hardware/software interface between the register and your program.

For your POS program you will need to have an intelligent cash drawer hardware interface that can accept an open signal and respond with a close signal as well.

I've seen drawers from M-S Cash Drawer and they fit what you are asking for.

It will provide you with the actual Visual Basic code, using their control, for connection to a USB port.  You can also get code for Parallel and Serial connections to the drawers.
