You can unplug the LAN cable... just kidding.

Depends, how is your configuration setup?

If it is a windows 2000 domain, you can disable internet access by ip or mac address etc...

OR  HKEY_CURRENT_USER\Software\Microsoft\Windows\CurrentVersion\Internet Settings]

Change the value of "ProxyEnable" and set it to "1". Change the value of "ProxyServer" and set it to an IP address and port that is invalid on your network such as "10.0.0.1:5555" (i.e. "IP:Port").
